There are entire books written on “how to be funny” but if you want my personal Cliff’s Notes version, just remember that it’s funny if it is not expected.  Therefore in order to be funny you simply need to do the exact opposite of what a normal and logical person would do.

If other average, ordinary guy would get defensive if someone accused him of being a virgin, you want to do the opposite.  If you want to be funny, simply embrace the accusation say something like “You’re right, I’m actually trying to set a new record and stay a virgin until I’m 41 years old.”  Not only is this funny, but it will come off as confident and it will set you apart from all the other defensive guys out there.
